II. Palmitic and stearic synthesis by an avocado supernatant system

چکیده
The synthesis of palmitic and stearic acids by soluble proteins from avocado mesocarp has been examined. Evidence suggests that palmitate is made de novo and stearate by elongation of a C16 precursor. While the synthesis of palmitate from malonyl CoA is arsenite insensitive, the elongation reaction from palmitate to stearate is arsenite sensitive. The pattern of synthesis of the two acids is neither dependent on the malonyl CoA/acetyl CoA ratio nor is altered by the addition of exogenous lipids and detergents. The reaction products are 20% thiol esters and 80% free fatty acids. When the incubation system was separated by gel filtration on Sephadex G-75 or G-150 columns two protein peaks were rapidly eluted. Over 60% of the C fatty acids were eluted with the protein peaks, the majority of the C fatty acids being in the first peak which contained only 10% of the total protein. The predominant acid of the first peak was stearate with 85% as the free acid. The second peak contained 30% thiol esters, including shorter-chain acids. It also contained fatty acid synthetase activity and [H]ACP. When [C-malonyl] CoA is added to a reaction mixture containing protein from this peak, palmitate is found exclusively. Thus, the de novo system is associated with this peak. The elongation system is no longer demonstrable with peaks 1 or 2. Unspecific binding of free fatty acids was negligible. [C]Palmityl-acyl carrier protein was elongated to stearate by an acetone powder of avocado supernatant fraction. It was a more effective precursor than [C]palmitic acid or [C-palmityl] CoA. From these results we conclude that avocado supernatant fraction contains a fatty acid synthetaseforming palmitate and an elongation system which forms stearate by malonyl CoA addition to a C16 precursor, possibly palmityl-ACP. The latter system is arsenite sensitive. *1 This research was supported by a NSF Grant GB-19733X. 2 Present address: Department of Biochemistry, University of Leeds, Leeds 2, England.

Triglyceride synthesis by the small-intestinal epithelium of the pig, sheep and chicken.
1. A comparative study was made of triglyceride synthesis by the intestinal epithelium of pigs, sheep and chickens. In pig and chicken tissue both the glycerol 3-phosphate and the monoglyceride pathway of triglyceride synthesis were operative, but the former pathway predominated in sheep tissue. 2. The fatty acid specificity of the glycerol 3-phosphate pathway was studied in pig and sheep total...
متن کاملMetabolic fate of oleic acid, palmitic acid and stearic acid in cultured hamster hepatocytes.
Unlike other saturated fatty acids, dietary stearic acid does not appear to raise plasma cholesterol. The reason for this remains to be established, although it appears that it must be related to inherent differences in the metabolism of the fatty acid. In the present study, we have looked at the metabolism of palmitic acid and stearic acid, in comparison with oleic acid, by cultured hamster he...
متن کاملMetabolic control analysis of de novo sunflower fatty acid biosynthesis.
We have obtained a simulation of the final steps of de novo fatty acid biosynthesis in sunflower control line RHA-274. For this simulation, we have used data from the evolution of fatty acids during seed formation and from the biochemical characterization of beta-keto-acyl-ACP synthetase II (FASII), stearoyl-ACP desaturase (SAD) and acyl-ACP thioesterase activities and the program GEPASI (based...
متن کاملRipening and in vitro retention of respiratory control by avocado and pear mitochondria.
The retention of respiratory control ("survival") by mitochondria held at 25 C was studied in relation to the ripening of two varieties of avocado (Persea americana Mill. var. ;Fuerte' and ;Hass') and one variety of pear (Pyrus communis. L. var. ;Bartlett') fruit. The survival of avocado mitochondria increased from 8 to 10 hours when isolated from unripe, preclimacteric fruit, to 48 hours when ...

A tandem Wittig approach has been employed for the synthesis of both (11S,9Z,12Z,15Z)- and (11R,9Z,12Z,15Z)-hydroxyoctadeca-9,12,15-trienoic acid (11-hydroxylinolenic acid, 11-HLA) from (R)-glyceraldehyde acetonide. From (11R)-HLA we have prepared the corresponding palmitic acid and stearic acid esters, mayolene-16 (1) and mayolene-18 (2), insect defensive compounds recently identified from Pie...
